iTunes 10.1 Download
The iTunes 10.1 update has been released for download, this is a required update to install the upcoming iOS 4.2 release on your iPhone, iPod touch, or iPad.
The release notes from Software Update are simple and as follows:

• Use AirPlay to instantly and wirelessly stream videos from iTunes to the all-new Apple TV.
• Sync with your iPhone, iPad, or iPod touch with iOS 4.2.
• Provides a number of important stability and performance improvements.
Download iTunes 10.1
The update available is for Mac OS X and Windows users. You have two options to download the update:
Software Update – You can use the Software Update feature from inside of iTunes, or you can go to Software Update from the Apple menu. Both will show you the download for iTunes 10.1.
Directly from Apple – If you go the Apple route, you don’t need to fill out your email, you can just click on the “Download Now” button. This will download an installer and you can then update iTunes yourself.

Any word on the iOS 4.2 release?
Apple has tiered the updates in typical fashion since each update is dependent on the next; Mac OS X update first, now iTunes, this means we will see iOS 4.2 early next week.
Don’t believe the rumor mongering elsewhere on the web, nobody has any insider tips, it’s all bologna.
I would agree with Greg here, this follows Apple’s usual release pattern.
